O padrão
Ponha not imediatamente depois do verbo:
I am not sure. She is not sure. They are not sure.
Nenhum auxiliar é emprestado, porque be consegue carregar a negação sozinho.
Duas maneiras de encurtar, uma lacuna
Para is e are há duas contrações, e as duas estão certas:
- he isn’t ou he*’s not***
- they aren’t ou they*’re not***
A diferença é de ênfase: he’s not ready carrega um pouco mais no not.
Para am há uma única opção. Amn’t não existe no inglês padrão, então a contração passa para o sujeito: I’m not.
Na pergunta a negação se parte
Numa pergunta negativa a contração fica com o verbo, mas a forma completa se divide:
- Isn’t she coming?
- Is she not coming? — mais formal, e um pouco mais seco.
no em vez de not
A negação também pode ser construída sobre o substantivo:
- There isn’t any milk.
- There is no milk.
As duas estão certas. A segunda é mais firme e mais comum na escrita.
Uma só negação, não duas
O português nega duas vezes sem nenhum problema: «não vi nada», «não falei com ninguém». O inglês aceita só uma:
- Não vi nada. → I didn’t see anything. / I saw nothing.
- Não há nenhum leite. → There isn’t any milk. / There is no milk.
Juntar as duas negações — there isn’t no milk — é transportar o padrão português, e em inglês é erro.

Erros a evitar
Errado: He doesn't be at home. certo: He isn't at home.
Be constrói a própria negativa. Acrescentar do ou does nunca está certo.
Errado: I amn't ready. certo: I'm not ready.
Não existe a contração amn't no inglês padrão. Encurte o sujeito em vez disso: I'm not.
Errado: They not are coming. certo: They aren't coming.
Not vem depois do verbo, nunca antes.
